I'm not totally sure who the largest clan RP guild is right now, for a long time it was either Clan Anarchist Syndicate or Mercury Dragons but as I understand those guilds have dwindled in size. As for roleplay pointers, the guide someone posted on the official roleplay forum probably says it better than I would:

Knowing how your character will speak and act in day to day situations is IMO much more important than having a fully fleshed out backstory. The backstory is a good and important thing, don't get me wrong, but people will notice and enjoy an interesting and consistent set of behaviors more often than they'll ask you for your personal history. You don't have to go hog wild, but some general ideas about how your character talks, what sort of decisions he/she will make in some common situations, and what kind of topics he/she might bring up in conversation will take you a long way.
My advice to you regarding guilds would be not to be too worried about the size of a roleplay guild, as it's not hard for a guild to sacrifice quality for quantity without even realizing it. Look for a guild with a playstyle that you're comfortable with, good quality roleplaying, nice people and enough members that you'll have people to interact with. Personally I'd give my guild, Whisper's Edge, top marks, and also recommend you check out the other guilds in the EndGame alliance. They're good folks, I'm glad to be hooked up with them.
