Mr Universe was an original concept that was already supposed to have been introduced to the series, and it would have been a lot more fleshed out, too.
Example: Know how Wash finished 2nd in his class in pilot school? Well, that's because Mr Universe was the one who finished first in that school, and not because he's a better pilot. He basically hacked all the school systems he was in and put his score to the top of the list in all of them. So Mr Universe's link to the rest of the crew is literally through Wash knowing (and having a former resentment-turned-respect for) him.
So the death of Mr. Universe and the death of Wash in the same film was a sort of tying-up of the loose ends that the second season would have had if the cancellation hadn't occurred. But yes, naturally, Mr Universe was never really introduced, so no-one really cared.
Any involvement that the characters of Wash or Book will have will be entirely back-story. Joss is many things, but he's not a completely blatant sell-out. He won't bring anyone back to life, but he might yet decide to do a separate spin-off to tell Book's back-story (and potentially introduce a new character to take over that hole). The best we can hope for with Wash is that he gives a few flashbacks from Zoe's perspective.
The only other factor that "could" happen, although unlikely, is a series and/or movie that retells the comic book interludes (the 6 months between the end of Objects in Space and the beginning of Serenity the Movie). That would obviously include Wash & Book, both alive and well, but it would be a bittersweet story, considering that we all know exactly how it's going to end. I don't think Joss likes that idea.
